**ADAPT-NK + bispecific NK cell engager targeting BCMA** is a combination immunotherapy for multiple myeloma consisting of ex vivo expanded adaptive NK cells and an affibody-derived bispecific NK cell engager that simultaneously binds CD16a on NK cells and B-cell maturation antigen (BCMA) on tumor cells. The bispecific engager redirects and activates NK cells to enhance cytotoxicity against BCMA-expressing plasma cells, potentially offering improved antitumor efficacy with reduced proinflammatory cytokine production compared to T-cell engagers. This approach leverages adaptive NK cells, which exhibit enhanced memory-like properties, in combination with the engager to boost NK cell persistence and tumor killing in relapsed/refractory multiple myeloma.[2]
