Adavosertib (AZD1775) + docetaxel is a combination therapy consisting of a WEE1 tyrosine kinase inhibitor and a taxane chemotherapy agent. Adavosertib regulates G2-M transition and maintains genomic stability, particularly in p53-deficient tumors which require DNA repair after genotoxic therapy. This combination exploits synthetic lethality in cancer cells, with adavosertib causing premature mitotic entry and genomic instability while docetaxel disrupts microtubule function. The combination has shown promising antitumor efficacy in clinical trials, particularly for head and neck squamous cell carcinoma (HNSCC).
