Overview

Adavosertib + olaparib is a combination of two small-molecule targeted therapies under investigation for the treatment of advanced and refractory solid tumors. Adavosertib (also known as AZD1775) is a first-in-class, selective inhibitor of Wee1-like protein kinase, which plays a key role in cell cycle regulation by inhibiting cyclin-dependent kinases 1 and 2. Inhibition of Wee1-like protein kinase disrupts the G2/M checkpoint, leading to increased DNA damage in cancer cells. Olaparib is an inhibitor of poly(ADP-ribose) polymerase (PARP), an enzyme involved in DNA repair; its inhibition leads to accumulation of DNA damage and cell death, particularly in tumor cells deficient in homologous recombination repair mechanisms (such as those with BRCA mutations). Preclinical data suggest that adavosertib enhances the antitumor effect of PARP inhibitors like olaparib by further compromising cancer cell DNA repair capacity. This combination has been evaluated primarily for patients with refractory or resistant solid tumors—including ovarian cancer—especially those with prior resistance to PARP inhibitors or platinum chemotherapy[1][2][3][5][6].
