**Adavosertib + paclitaxel** is a combination regimen used in oncology clinical trials. Adavosertib is a small molecule inhibitor of the serine/threonine-protein kinase WEE1, often used under code names AZD1775 or MK-1775. Paclitaxel is a well-established microtubule-stabilizing antineoplastic small molecule. This combination is primarily being investigated for the treatment of advanced solid tumors, especially platinum-sensitive, TP53-mutated ovarian cancer and other malignancies. Adavosertib sensitizes tumor cells to DNA damage by abrogating cell cycle checkpoints, especially in TP53-deficient cells, thereby enhancing the cytotoxic effects of paclitaxel. Combination regimens typically include carboplatin, but there are regimens with adavosertib plus paclitaxel alone being tested in various solid tumors[1][2][3][5][7].
