Overview

This is an investigational combination regimen consisting of **adavosertib** (a selective, oral small-molecule inhibitor of WEE1 kinase), **paclitaxel** (an anti-mitotic chemotherapy agent), and **carboplatin** (a platinum-based chemotherapeutic agent). Adavosertib inhibits WEE1, a key regulator of the G2/M checkpoint in the cell cycle, thereby forcing cells with DNA damage (such as p53-mutant tumor cells) to enter mitosis prematurely, leading to cell death. Paclitaxel disrupts microtubule dynamics, preventing cell division, while carboplatin forms DNA crosslinks that induce apoptosis. This combination has been studied primarily for advanced solid tumors—particularly platinum-sensitive ovarian, fallopian tube, and primary peritoneal cancers with p53 mutations, but also in other advanced solid tumors, including squamous cell lung carcinoma. It is not an approved regimen and remains investigational[1][2][3][5].
