AdCh63 AMA1 + AMA1-C1 is a combination malaria vaccine candidate targeting the blood-stage of Plasmodium falciparum infection. The combination consists of two components: - **AdCh63 AMA1**: A non-replicating simian adenovirus vector (chimpanzee adenovirus serotype 63) genetically engineered to express the apical membrane antigen 1 (AMA1) from P. falciparum. This viral vector is designed to induce strong cellular and humoral immune responses against the malaria parasite[3][5][7]. - **AMA1-C1**: A recombinant protein vaccine composed of a mixture of two synthetic forms of the P. falciparum apical membrane antigen 1 (from different strains), formulated with Alhydrogel (an aluminum-based adjuvant). It may also be administered with CPG 7909, an oligodeoxynucleotide adjuvant that acts as a TLR9 agonist to further enhance immunogenicity[6]. The rationale for combining these modalities is to leverage both robust T-cell priming from the viral vector and potent antibody responses from the protein/adjuvant formulation, aiming for improved immunogenicity and efficacy over either approach alone[3][5][6]. The primary indication under investigation is prevention of malaria caused by P. falciparum.
