AdCh63 AMA1 + MVA AMA1 + AMA1-C1 + CPG 7909 is a multi-component, heterologous prime-boost malaria vaccine regimen developed by the University of Oxford. The regimen targets the blood-stage of Plasmodium falciparum infection by inducing immune responses against the Apical Membrane Antigen 1 (AMA1), a protein essential for merozoite invasion of erythrocytes. It utilizes two viral vector vaccines—a chimpanzee adenovirus (AdCh63) and a modified vaccinia virus Ankara (MVA)—both engineered to express the AMA1 antigen. These are combined with a recombinant protein component, AMA1-C1, which contains AMA1 sequences from the FVO and 3D7 clones of P. falciparum. The protein is formulated with Alhydrogel and the TLR9 agonist adjuvant CPG 7909 to enhance immunogenicity. This mixed-modality approach is designed to elicit potent cellular and humoral immune responses to block malaria infection.
