Adebrelimab + dalpiciclib is an investigational combination therapy comprising two distinct agents: adebrelimab, a humanized monoclonal antibody targeting programmed death-ligand 1 (PD-L1), and dalpiciclib, a selective small molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6). Adebrelimab acts as an immune checkpoint inhibitor by blocking PD-L1, thereby enhancing anti-tumor immune responses. Dalpiciclib inhibits CDK4/6-mediated cell cycle progression, leading to cell cycle arrest in tumor cells with dysregulated CDKN2A or amplified CDK4/6 activity. The combination is being studied for synergistic anti-tumor effects in various cancers, including hormone receptor-positive/human epidermal growth factor receptor 2-negative (HR+/HER2-) breast cancer and HPV-negative locally advanced head and neck squamous cell carcinoma (LAHNSCC)[3][4][6]. Both drugs are developed by Jiangsu Hengrui Medicine.
