Adebrelimab + SHR-8068 + chemotherapy is a combination immunotherapy and cytotoxic regimen developed by Suzhou Suncadia Biopharmaceuticals (a subsidiary of Jiangsu Hengrui Pharmaceuticals) for the treatment of locally advanced resectable rectal cancer. The regimen consists of adebrelimab, a humanized IgG4 monoclonal antibody targeting the Programmed Cell Death 1 Ligand 1 (PD-L1), and SHR-8068, a monoclonal antibody targeting Cytotoxic T-Lymphocyte Antigen 4 (CTLA-4). These are combined with standard chemoradiotherapy in a perioperative setting. By simultaneously blocking the PD-L1/PD-1 and CTLA-4 pathways, the therapy aims to overcome immune evasion mechanisms and synergize with the immunogenic cell death induced by chemotherapy and radiation, potentially increasing the rate of pathological complete response (pCR) in rectal cancer patients.
