Adebrelimab + TP + FP is a combination immunotherapy-chemotherapy regimen currently being evaluated in Phase II clinical trials for the treatment of locally advanced unresectable esophageal squamous cell carcinoma (ESCC). The regimen consists of adebrelimab (SHR-1316), a humanized IgG4 monoclonal antibody targeting programmed cell death-ligand 1 (PD-L1), combined with either a taxane-platinum (TP) doublet (paclitaxel or nab-paclitaxel plus carboplatin) or a fluoropyrimidine-platinum (FP) doublet (5-fluorouracil plus cisplatin). In the clinical trial setting (NCT05463107), patients receive induction therapy with adebrelimab and chemotherapy, followed by PET-CT-guided concurrent chemoradiotherapy (CCRT) and maintenance adebrelimab. The goal of this approach is to improve the conversion rate to resectability or achieve better local control in unresectable ESCC by leveraging the synergistic effects of PD-L1 inhibition and cytotoxic chemotherapy.
