Overview

Adefovir + lamivudine is a combination of two antiviral drugs used primarily for the treatment of chronic hepatitis B virus (HBV) infection, especially in cases where resistance to monotherapy has developed. Adefovir is an acyclic nucleotide analog of adenosine monophosphate that inhibits HBV DNA polymerase (reverse transcriptase) by competing with deoxyadenosine triphosphate and causing DNA chain termination after incorporation into viral DNA[2][6]. Lamivudine is a nucleoside reverse transcriptase inhibitor (NRTI) that acts as a synthetic nucleoside analogue, phosphorylated intracellularly to its active metabolite, which inhibits both HIV reverse transcriptase and HBV polymerase by causing premature termination of the viral DNA chain[3][4][5]. The combination provides additive anti-HBV activity and may be used when resistance to either agent alone occurs.
