Overview

Adefovir dipivoxil + polyene phosphatidylcholine is a combination therapy investigated primarily for the treatment of chronic hepatitis B. Adefovir dipivoxil is an oral prodrug of adefovir, an acyclic nucleotide analog of adenosine monophosphate. After conversion to its active metabolite, adefovir diphosphate, it inhibits hepatitis B virus (HBV) DNA polymerase (reverse transcriptase) by competing with deoxyadenosine triphosphate and causing DNA chain termination upon incorporation into viral DNA[6]. Polyene phosphatidylcholine (PPC) is used in China as a hepatoprotective agent for various liver diseases and drug-induced liver injury; it acts by stabilizing cell membranes and promoting hepatic regeneration[5]. The rationale for combining these agents is to enhance antiviral efficacy while potentially reducing hepatotoxicity associated with long-term antiviral therapy.
