Adenosine 5'-[gamma-thio]triphosphate is a synthetic nucleoside triphosphate analogue of ATP in which one of the oxygens on the terminal (gamma) phosphate group is replaced by sulfur. This modification makes it resistant to hydrolysis by many enzymes, allowing it to act as a non-hydrolyzable or slowly hydrolyzable substitute for ATP in biochemical and physiological studies. It is widely used as a research tool to study processes involving ATP binding and hydrolysis, including kinase reactions (where it yields thiophosphorylated proteins that are resistant to protein phosphatases), purinergic signaling (as an agonist at P2 receptors), modulation of Ca2+-activated K+ channels, and investigations into cell signaling pathways, enzyme kinetics, neurotransmitter release, and proteasome function. It is not approved for clinical use but has broad application in molecular biology, neuroscience research, and drug development[1][2][4].
