Overview

AdFITC(E2)-CAR T-cells are an adaptor-based chimeric antigen receptor (CAR) T-cell therapy designed for modular and tunable tumor targeting. These genetically engineered T-cells express a second-generation CAR consisting of an anti-fluorescein (FITC) single-chain variable fragment (scFv) derived from the E2 antibody clone, a CD8α hinge, a 4-1BB co-stimulatory domain, and a CD3ζ activation domain. Unlike traditional CAR T-cells that target tumor antigens directly, AdFITC(E2)-CAR T-cells recognize FITC-labeled adaptor molecules (such as FITC-conjugated diabodies or peptides like Octo-Fluo) that bridge the CAR T-cell to specific tumor-associated antigens. This approach allows for the targeting of multiple antigens (e.g., SSTR2, CD33, CD117) and provides a safety mechanism where CAR T-cell activity can be controlled by the administration of the adaptor. Research has explored its application in gastro-entero-pancreatic neuroendocrine tumors (GEP-NETs), acute myeloid leukemia (AML), and various solid tumors.
