Overview

ADG106 + paclitaxel is an investigational combination therapy being studied primarily in HER2-negative breast cancer. ADG106 is a ligand-blocking agonistic monoclonal antibody targeting CD137 (also known as 4-1BB), a costimulatory receptor expressed on activated T cells and natural killer (NK) cells. By activating CD137, ADG106 enhances anti-tumor immune responses through immune cell activation and cytokine modulation. Paclitaxel is a well-established chemotherapeutic agent that stabilizes microtubules, inhibiting cell division and inducing apoptosis in rapidly dividing tumor cells. The combination aims to leverage the immunostimulatory effects of ADG106 with the cytotoxic activity of paclitaxel to improve neoadjuvant treatment outcomes in patients with HER2-negative breast cancer[1][2][5].
