ADG106 + toripalimab

ADG106 + toripalimab is an investigational combination immunotherapy being developed by Adagene in collaboration with Junshi Biosciences. The regimen consists of ADG106, a fully human ligand-blocking agonistic monoclonal antibody targeting CD137 (4-1BB), and toripalimab, a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. ADG106 is engineered using Adagene's NEObody™ technology to bind a unique epitope of CD137, providing a potent costimulatory signal to activated T cells and natural killer cells. Toripalimab acts as a checkpoint inhibitor by blocking the inhibitory PD-1/PD-L1 pathway, thereby restoring anti-tumor immune responses. This combination is designed to synergistically enhance the immune system's ability to target cancer cells and is being evaluated in Phase 1b/2 clinical trials (such as NCT04775680) for patients with advanced solid tumors and relapsed or refractory non-Hodgkin lymphoma.
