ADG126 + ADG106 is a combination of two investigational monoclonal antibodies developed by Adagene for the treatment of advanced or metastatic solid tumors. - **ADG126** is a fully human anti-CTLA-4 IgG1 antibody prodrug engineered with Adagene's SAFEbody technology, which masks the antibody binding site to limit off-tumor toxicity and is preferentially activated in the tumor microenvironment. Upon activation, it blocks Cytotoxic T lymphocyte-associated protein 4 (CTLA-4) and depletes regulatory T cells via enhanced antibody-dependent cellular cytotoxicity (ADCC), aiming to improve safety and efficacy over existing Cytotoxic T lymphocyte-associated protein 4 (CTLA-4) therapies[1][3][6][7]. - **ADG106** is a fully human agonistic IgG4 monoclonal antibody targeting Tumor necrosis factor receptor superfamily member 9 (CD137, also known as 4-1BB), designed to stimulate immune responses against cancer by activating T cells through Tumor necrosis factor receptor superfamily member 9 (CD137) engagement. It acts as a ligand-blocking agonist, promoting T-cell proliferation and survival while blocking reverse signaling of the Tumor necrosis factor receptor superfamily member 9 (CD137) ligand[5][8]. The combination leverages dual immune checkpoint modulation—blocking immunosuppressive pathways (Cytotoxic T lymphocyte-associated protein 4 (CTLA-4)) while stimulating co-stimulatory signals (Tumor necrosis factor receptor superfamily member 9 (CD137))—to enhance antitumor immunity. Both agents are being evaluated in early-phase clinical trials for patients with advanced/metastatic solid tumors[2].
