Overview

ADG126 + pembrolizumab is a combination immunotherapy consisting of ADG126, a fully human anti-CTLA-4 monoclonal antibody utilizing a protease-cleavable masking (SAFEbody) technology for tumor-selective activation, and pembrolizumab, a PD-1 receptor-blocking humanized IgG4 monoclonal antibody. ADG126 is preferentially activated in the tumor microenvironment, leading to enhanced T cell priming via partial CTLA-4 ligand blockade and potent regulatory T cell depletion through antibody-dependent cellular cytotoxicity (ADCC). Pembrolizumab inhibits the PD-1 pathway to restore and sustain anti-tumor immune responses. The combination is being developed for advanced/metastatic solid tumors, particularly metastatic microsatellite stable colorectal cancer (MSS CRC) without liver metastasis. Early-phase clinical trials demonstrate manageable toxicity and encouraging efficacy, with improved response rates relative to conventional CTLA-4/PD-1 combinations[1][2][3][4][5].
