Overview

ADG126 + toripalimab is an investigational combination therapy comprising two monoclonal antibodies: **ADG126**, a fully human IgG1 anti-CTLA-4 SAFEbody with a masking peptide that is selectively activated in the tumor microenvironment (TME), and **toripalimab**, an anti-PD-1 immune checkpoint inhibitor. ADG126, once activated in the TME, binds a unique epitope of CTLA-4, allowing for enhanced T-cell activation and selective depletion of immunosuppressive regulatory T cells (Tregs) via antibody-dependent cellular cytotoxicity within tumors. Toripalimab blocks PD-1, restoring effector T-cell anti-tumor activity. The combination is designed to maximize anti-tumor immune responses with reduced systemic toxicity compared to conventional anti-CTLA-4 therapy. Primary clinical development is in advanced/metastatic solid tumors[1][3][5].
