Aanastra is developing ADGN-121 and ADGN-122, which are peptide-RNA nanoparticle therapeutics designed to selectively delete or silence oncogenic KRAS mutations. ADGN-121 specifically targets the KRAS G12D mutation, while ADGN-122 targets the KRAS G12V mutation. These compounds are part of Aanastra's ONCOEDIT platform (Oncogene Editing/Deletion), which utilizes gene editing or gene silencing approaches delivered via the company's proprietary PEP-NP (peptide nanoparticle) technology to target tumor cells. The therapeutics consist of RNA-based molecules combined with proprietary targeting peptides that enable in vivo delivery to tumors. Both compounds are currently in preclinical development and are being investigated for the treatment of cancers driven by KRAS mutations, including colorectal cancer, lung cancer, and pancreatic cancer. The mechanism involves the selective deletion of mutant KRAS genes to overcome acquired resistance to small molecule KRAS inhibitors.
