This is a research-stage injectable thermosensitive composite hydrogel designed for the treatment of knee osteoarthritis. It consists of adipose-derived mesenchymal stem cell (AD-MSC) supernatant, which contains a variety of paracrine factors and bioactive molecules, encapsulated within a matrix of Pluronic F-127 (PF-127) and hyaluronic acid (HA). The formulation acts as a cell-free therapeutic, leveraging the regenerative and anti-inflammatory properties of MSC secretions without the complexities of live cell transplantation. The hydrogel remains in a liquid state at low temperatures (4°C), allowing for easy intra-articular injection, and rapidly transitions into a stable gel at physiological temperature (37°C). This transition provides localized retention and sustained release of the bioactive factors, which have been shown in preclinical models to reduce proinflammatory cytokines (such as IL-1β and TNF-α), decrease cartilage degradation markers (like CTX-II), and preserve joint architecture.
