A combination therapy consisting of ado-trastuzumab emtansine (T-DM1), an antibody-drug conjugate targeting HER2-positive cancer cells, and ribociclib, a CDK4/6 inhibitor. This combination is being investigated for the treatment of HER2-positive metastatic breast cancer in patients previously treated with trastuzumab and taxanes. Preclinical evidence suggests synergy between CDK4/6 inhibitors and anti-HER2 therapies.
