Overview

ADP-A2M4 + pembrolizumab is an investigational combination therapy consisting of a T cell receptor (TCR) engineered cell therapy (ADP-A2M4, also known as afamitresgene autoleucel) that specifically targets the cancer testis antigen MAGE-A4, and pembrolizumab, a monoclonal antibody checkpoint inhibitor targeting the PD-1 pathway. ADP-A2M4 is developed to treat cancers expressing MAGE-A4, particularly in HLA-A*02 positive patients. The rationale for combining ADP-A2M4 with pembrolizumab is to potentiate anti-tumor immune responses by leveraging direct tumor recognition by engineered T cells and simultaneous blockade of immune suppression via PD-1 inhibition. The combination is being clinically investigated in advanced and metastatic cancers, particularly head and neck cancer, with early studies suggesting enhanced anti-tumor activity and a tolerable safety profile[1][2][4].
