ADP-A2M4CD8 + nivolumab is an investigational combination therapy comprising ADP-A2M4CD8, a next-generation autologous T-cell receptor (TCR) T-cell therapy, and nivolumab, a programmed death 1 (PD-1) checkpoint inhibitor. ADP-A2M4CD8 uses genetically modified autologous T-cells engineered to express a high-affinity T-cell receptor specific for the MAGE-A4 cancer testis antigen, paired with a CD8α co-receptor to enhance CD4+ T-cell function and cytotoxicity. This product enables robust immune recognition and attack of HLA-A*02-positive, MAGE-A4-expressing solid tumors. Nivolumab, a human monoclonal antibody, blocks the PD-1 receptor to further potentiate anti-tumor T-cell activity by preventing inhibitory signaling. The combination is being developed for advanced or metastatic solid tumors, with clinical trials focusing on platinum-resistant ovarian cancer, urothelial cancer, and head and neck cancer. The program is being advanced by Adaptimmune and is currently in phase 2 development1[1][2][5][7][9].
