Overview

ADP-A2M4CD8 + pembrolizumab is a combination therapy comprised of two components: - **ADP-A2M4CD8** is an autologous, genetically engineered T-cell therapy that expresses an affinity-enhanced T-cell receptor (TCR) targeting the cancer testis antigen **MAGE-A4** and co-expresses the CD8α co-receptor. This design enhances recognition and cytotoxicity against MAGE-A4 positive tumor cells in HLA-A*02 positive patients. It is developed using Adaptimmune's SPEAR (Specific Peptide Enhanced Affinity Receptor) T-cell platform. The therapy is administered following lymphodepleting chemotherapy. - **Pembrolizumab** is a small molecule monoclonal antibody checkpoint inhibitor targeting **programmed cell death protein 1 (PD-1)**, thereby preventing immune suppression and enhancing T cell-mediated antitumor activity. This combination aims to enhance the antitumor activity of ADP-A2M4CD8 T cells via the immunomodulatory effects of pembrolizumab, particularly in solid tumors expressing MAGE-A4 such as ovarian, bladder, and head and neck cancers. Clinical studies are ongoing to assess efficacy and safety in these indications.
