ADT + EBRT is a therapeutic combination consisting of Androgen Deprivation Therapy (ADT) and External Beam Radiation Therapy (EBRT). In the DARIUS (AFU-GETUG P15) phase II clinical trial sponsored by Institut Bergonié, this combination serves as the standard-of-care control arm for patients with unfavorable intermediate-risk prostate cancer. ADT involves the use of LHRH agonists or antagonists to suppress testosterone levels, thereby depriving prostate cancer cells of the androgens required for growth. EBRT utilizes high-energy radiation beams to target and destroy cancer cells in the prostate. The trial compares this standard regimen against an experimental arm of darolutamide plus EBRT to assess if the androgen receptor antagonist can provide comparable efficacy with a potentially improved safety profile and quality of life.
