**ADU-1805 + pembrolizumab** is a combination therapy under investigation for advanced solid tumors. ADU-1805 is a humanized monoclonal IgG2 antibody that targets all known human SIRPα alleles and blocks the interaction between SIRPα and CD47, thereby promoting phagocytosis by macrophages and enhancing neutrophil trogocytosis. Unlike CD47-targeting agents, ADU-1805 does not interfere with T-cell activation. Pembrolizumab is a monoclonal antibody checkpoint inhibitor against PD-1. The combination aims to enhance anti-tumor immunity by both stimulating innate immune phagocytosis and releasing adaptive immune suppression. The combination is currently being evaluated in first-in-human, open-label, multicenter, multi-arm phase 1 dose escalation studies, including arms for ADU-1805 monotherapy and ADU-1805 plus pembrolizumab[1][3][5][6][9].
