Overview

ADU-S100 + pembrolizumab is an investigational combination therapy for recurrent or metastatic head and neck squamous cell carcinoma (HNSCC). ADU-S100 (MIW815) is a synthetic cyclic dinucleotide that acts as an agonist of the stimulator of interferon genes (STING) pathway, leading to innate immune activation and enhanced anti-tumor immunity. Pembrolizumab is a monoclonal antibody that blocks the programmed cell death protein 1 (PD-1) receptor, restoring T-cell activity against tumor cells. The combination aims to synergistically boost immune responses against cancer by activating both innate (via STING) and adaptive (via PD-1 inhibition) immune pathways[1][3][5]. ADU-S100 is administered by intratumoral injection; pembrolizumab is given as an intravenous infusion. The development program was sponsored by Chinook Therapeutics (formerly Aduro Biotech), and clinical trials involved a research partnership with Novartis. Development status for this combination in HNSCC is Phase 2, but trials have been discontinued due to lack of substantial antitumor activity[5][3].
