AdVTMM2 + dendritic cell is an investigational cancer immunotherapy that combines autologous human dendritic cells (DCs) with a recombinant adenoviral vector (AdVTMM2) engineered to express three full-length melanoma-associated antigens: tyrosinase, MART-1, and MAGE-A6. The therapy involves transducing patient-derived DCs ex vivo with the E1/E3-deleted adenovirus encoding these antigens. The resulting antigen-loaded DCs are then administered to the patient as a vaccine. This approach aims to activate both CD8+ and CD4+ T cells against multiple melanoma antigens simultaneously, promoting polyclonal anti-tumor immunity and enhancing both adaptive and innate immune responses—including natural killer (NK) cell activation. Preclinical studies have shown that this multi-antigen strategy leads to broader T-cell activation compared to single-antigen approaches and may result in improved clinical outcomes for patients with melanoma[1].
