**ADXS-NEO + pembrolizumab** is a combination investigational immunotherapy regimen being studied primarily in advanced/metastatic solid tumors, especially non-small cell lung cancer (NSCLC). ADXS-NEO is a personalized, off-the-shelf cancer vaccine platform using live attenuated *Listeria monocytogenes* vectors, bioengineered to encode up to 40 tumor-specific neoantigens unique to each patient, aiming to stimulate potent antigen-specific T-cell responses against tumor cells. Pembrolizumab is a monoclonal antibody immune checkpoint inhibitor targeting PD-1, restoring and enhancing antitumor immune responses by blocking PD-1-mediated immunosuppression. The combination is designed to boost immune recognition of tumor neoantigens and to overcome resistance/to enhance responsiveness to checkpoint inhibition.
