AEG35156 + gemcitabine is a drug combination consisting of an **antisense oligonucleotide (AEG35156)** targeting the X-linked inhibitor of apoptosis protein (XIAP) mRNA, and **gemcitabine**, a nucleoside analog antimetabolite used as chemotherapy. AEG35156 reduces XIAP expression, thereby promoting apoptosis and potentially sensitizing cancer cells to chemotherapy. Preclinical studies demonstrated potent activity in pancreatic ductal adenocarcinoma (PDA). In a phase I clinical trial in metastatic PDA, the combination aimed to establish safety, maximum tolerated dose, and preliminary efficacy. The combination did not demonstrate significant antitumor activity in advanced PDA.
