Drug intelligence / Profile preview
AER001 + AER002 is an investigational combination of two long-acting, fully human IgG1 monoclonal antibodies, AER001 (also known as P5C3) and AER002 (also known as P2G3), originally discovered by Lausanne University Hospital (CHUV) and Ecole Polytechnique Fédérale de Lausanne (EPFL) and developed by Aerium Therapeutics. Both antibodies target non-overlapping epitopes on the receptor-binding domain (RBD) of the SARS-CoV-2 Spike protein, competing with angiotensin-converting enzyme 2 (ACE2) to block viral attachment and entry into host cells. The antibodies are engineered with the LS modification (Met428Leu/Asn434Ser) to extend their in vivo half-life. The combination was evaluated in a Phase 1 trial for the pre-exposure prophylaxis and treatment of COVID-19, particularly in immunocompromised individuals. However, development was discontinued after Aerium Therapeutics ceased operations in July 2024 due to the loss of neutralization activity against emerging Omicron subvariants.
