Overview

Afatinib + anti-EGF vaccine is a combination therapy consisting of afatinib, an oral irreversible tyrosine kinase inhibitor (TKI) targeting the ErbB family of receptors (including EGFR), and an anti-epidermal growth factor (EGF) vaccine designed to induce antibodies against EGF. This approach aims to both inhibit EGFR signaling directly via TKI activity and reduce ligand-driven activation by neutralizing circulating EGF through vaccination-induced antibodies. The combination has been studied in advanced non-small cell lung cancer (NSCLC) patients with activating EGFR mutations, showing promising efficacy, sustained immunogenic response, and manageable safety profile in early-phase clinical trials[1][2][3][5]. The rationale is that dual blockade—by inhibiting the receptor with afatinib and depleting its ligand with the vaccine—may enhance antitumor effects and delay resistance compared to TKI monotherapy.
