This combination therapy consists of the irreversible ErbB family blocker afatinib, the anti-VEGF monoclonal antibody bevacizumab, and the folate antimetabolite pemetrexed. It is being investigated primarily by Shandong Cancer Hospital for the treatment of patients with advanced lung adenocarcinoma who have progressed after treatment with first-generation EGFR tyrosine kinase inhibitors (TKIs). The regimen aims to overcome resistance by simultaneously inhibiting multiple ErbB receptors (EGFR, HER2, and ErbB4), blocking tumor angiogenesis via VEGF inhibition, and providing cytotoxic chemotherapy. This triplet approach is designed to address the heterogeneity of resistance mechanisms in non-small cell lung cancer (NSCLC).
