Afatinib + nivolumab is an investigational combination regimen involving a small molecule tyrosine kinase inhibitor (afatinib) and an immunotherapeutic monoclonal antibody (nivolumab). Afatinib is an irreversible inhibitor of the ErbB family of receptor tyrosine kinases (specifically EGFR, HER2, HER4), approved primarily for the treatment of EGFR mutation-positive non-small cell lung cancer (NSCLC). Nivolumab is a fully human IgG4 monoclonal antibody that inhibits the immune checkpoint receptor PD-1, reactivating anti-tumor immune responses. The combination is in early-stage clinical trials for solid tumors such as NSCLC, aiming to target tumor growth through dual inhibition of oncogenic signaling and immune evasion mechanisms.
