Afatinib + paclitaxel is a combination therapy used in oncology, particularly for patients with advanced solid tumors and metastatic breast cancer who have failed prior HER2-targeted therapies. Afatinib is an irreversible inhibitor of the ErbB family of tyrosine kinases, including EGFR (ErbB1), HER2 (ErbB2), and HER4 (ErbB4). It blocks intracellular signaling pathways involved in tumor cell proliferation and survival. Paclitaxel is a microtubule-stabilizing agent that inhibits cell division by promoting tubulin polymerization and preventing microtubule disassembly.
