Afatinib + panitumumab is a combination therapy that targets the epidermal growth factor receptor (EGFR) through dual inhibition mechanisms. Afatinib is an ErbB family blocker that irreversibly inhibits signaling from EGFR (ErbB1), HER2 (ErbB2), HER4 (ErbB4), and all relevant ErbB family dimers. Panitumumab is a fully human IgG2 monoclonal antibody that binds specifically to the extracellular domain of EGFR, preventing binding of endogenous ligands such as epidermal growth factor (EGF) or transforming growth factor-alpha. This combination has been studied for overcoming resistance in EGFR-mutated non-small-cell lung cancer.
