A combination of afatinib, an irreversible ErbB family blocker that covalently binds to the kinase domains of epidermal growth factor receptor (EGFR) and human EGFRs (HER), and sirolimus, an mTOR inhibitor. This combination was investigated in a phase Ib clinical trial for patients with EGFR mutation-positive non-small-cell lung cancer (NSCLC) and/or disease progression following prior erlotinib/gefitinib treatment. It was studied as a potential approach to reverse acquired resistance to EGFR tyrosine kinase inhibitors (EGFR-TKIs).
