Overview

Aflibercept + gemcitabine is a combination therapy that pairs aflibercept, a recombinant fusion protein acting as a decoy receptor for vascular endothelial growth factors (VEGF-A, VEGF-B, and placental growth factor), with gemcitabine, a nucleoside analog chemotherapy. This combination has been clinically investigated in advanced solid tumors, especially metastatic pancreatic cancer, aiming to inhibit tumor angiogenesis and tumor cell proliferation. While preclinical studies showed enhanced antitumor efficacy compared to monotherapy, clinical trials in pancreatic cancer did not demonstrate improved overall survival. The combination has been tested primarily by intravenous administration.
