AFM13 + AB-101 is a combination investigational cancer immunotherapy currently in phase 2 clinical studies for the treatment of relapsed or refractory (R/R) classical Hodgkin lymphoma (cHL) and CD30-positive peripheral T-cell lymphoma (PTCL). AFM13 is a tetravalent, bispecific innate cell engager (ICE) monoclonal antibody designed to bind CD30 on tumor cells and CD16A (FcγRIIIa) on natural killer (NK) cells, bridging NK cells to CD30-positive malignant cells and leading to potent antibody-dependent cellular cytotoxicity (ADCC). AB-101 is a non-genetically modified, cryopreserved, allogeneic, cord blood-derived NK cell product optimized for combinational use with monoclonal antibodies such as AFM13; it is pre-selected for high-affinity CD16A and the KIR-B haplotype to enhance NK cell activity and ADCC. The combination leverages AFM13's tumor cell targeting with enhanced cytotoxicity through adoptive transfer of AB-101, providing a scalable, off-the-shelf approach to targeting CD30+ blood cancers[1][3][5][7].
