Overview

AFM13 + pembrolizumab is a combination therapy under clinical investigation for relapsed or refractory Hodgkin lymphoma. **AFM13** is a bispecific, tetravalent innate immune cell engager antibody targeting CD30 on lymphoma cells and CD16A on natural killer cells and macrophages, thus promoting tumor cell killing through innate immunity. **Pembrolizumab** (brand name Keytruda; code name MK-3475) is a monoclonal antibody acting as an immune checkpoint inhibitor by targeting programmed death-1 (PD-1) to release T-cell mediated antitumor activity. The combination leverages both innate (NK/macrophage) and adaptive (T cell) immune responses for enhanced efficacy. In phase 1b studies, the therapy was well-tolerated with high overall and complete metabolic response rates, especially at the highest dose cohort[1][2][3][5].
