**afuresertib + sintilimab + docetaxel** is a combination therapy under clinical investigation for the treatment of solid tumors resistant to prior anti-PD-1/PD-L1 therapy, including non-small cell lung cancer (NSCLC), gastric cancer/gastroesophageal junction cancer, esophageal cancer, cervical cancer, and endometrial cancer. - **Afuresertib** is an orally available, selective small molecule inhibitor of AKT, a kinase in the PI3K/AKT pathway involved in cell survival and proliferation. - **Sintilimab** is a human IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, acting as an immune checkpoint inhibitor to restore T-cell mediated anti-tumor responses. - **Docetaxel** is a cytotoxic chemotherapy agent that inhibits microtubule depolymerization, leading to cell cycle arrest and apoptosis. The combination aims to overcome resistance to immune checkpoint blockade by combining immunotherapy with pathway inhibition and cytotoxic chemotherapy.
