Overview

This is a combination regimen that includes the following three agents: - **Afuresertib**, an oral, ATP-competitive, reversible small molecule inhibitor of AKT kinases (AKT1, AKT2, AKT3), targeting the PI3K/AKT pathway, with anti-proliferative and pro-apoptotic effects in cancer cells; - **Sintilimab**, a recombinant humanized IgG4 monoclonal antibody that acts as a immune checkpoint inhibitor by binding to the PD-1 receptor on T-cells, blocking PD-1/PD-L1 interaction, and enhancing anti-tumor immune responses; - **Nab-paclitaxel**, an albumin-bound formulation of the chemotherapy drug paclitaxel, a microtubule inhibitor that disrupts cell division and induces apoptosis in rapidly proliferating tumor cells. This triple combination is under investigation for the treatment of specific solid tumors, particularly in patients who have progressed after prior anti-PD-1/PD-L1 treatments (such as immune checkpoint inhibitors or chemotherapy). Indications under study include gynecologic cancers (notably cervical and endometrial), as well as other solid tumors resistant to standard therapies. Phase I/II trials have shown promising anti-tumor activity and tolerable safety in these settings[1][3][7].
