AG-707 + QS-21 is an investigational therapeutic vaccine combination designed to treat herpes simplex virus-2 (HSV-2, which causes genital herpes). AG-707 consists of recombinant human heat shock protein-70 complexed with 32 distinct synthetic peptides derived from the HSV-2 proteome, aiming to elicit broad immune responses against multiple viral antigens. This construct is formulated with QS-21, a purified saponin-based adjuvant from *Quillaja saponaria* that enhances immune responses by promoting both CD4+ and CD8+ T cell activation. The combination has demonstrated robust, statistically significant activation of both cellular (T-cell) arms of immune response in phase 1 trials for HSV-2. AG-707 + QS-21 has been developed by Antigenics and was tested clinically for genital herpes, with the vaccine being well tolerated and mostly associated with local injection site reactions.
