Overview

AGEN2373 + botensilimab is a combination of two investigational monoclonal antibodies for immunotherapy of advanced solid tumors, including melanoma. **AGEN2373** is a conditionally active CD137 (4-1BB) agonist antibody, engineered to selectively enhance tumor immunity and minimize off-target toxicity by binding a unique epitope on effector T and NK cells. **Botensilimab** (also known as AGEN1181) is an Fc-enhanced CTLA-4 antagonist antibody designed to block the inhibitory CTLA-4 receptor, promote immune cell activation, and enhance tumor killing. Combined, these agents aim to synergistically boost anti-tumor immune responses, especially in patients resistant to prior PD-(L)1 therapies. This combination is under clinical investigation by Agenus, with early evidence of safety and antitumor activity in heavily pretreated populations[1][3][6][7].
