AGENT-797 + nivolumab is a cellular immunotherapy combination comprising AGENT-797, an allogeneic invariant natural killer T (iNKT) cell therapy, and nivolumab, a monoclonal antibody immune checkpoint inhibitor targeting PD-1. AGENT-797 is an off-the-shelf iNKT cell product that provides both direct cytotoxic anti-tumor effects (via killing tumor cells and modulating the tumor microenvironment) and broad immune system activation by recognition of lipid antigens presented on CD1d, secretion of cytokines (such as IFN-γ), and enhancement of cytotoxic lymphocyte infiltration. Nivolumab blocks the PD-1 receptor, preventing its inhibitory interaction with PD-L1 on tumor cells, thereby restoring and augmenting T-cell–mediated immune responses against cancer. The combination has demonstrated clinical activity in PD-1–refractory cancers, such as gastric adenocarcinoma, with increased tumor infiltration by cytotoxic lymphocytes and durable tumor regression[1][2][3][4][7].
