Overview

AGENT-797 + pembrolizumab is an investigational combination therapy comprised of AGENT-797, an allogeneic invariant natural killer T (iNKT) cell therapy, and pembrolizumab, a programmed death-1 (PD-1) immune checkpoint inhibitor. AGENT-797 is designed to activate both innate and adaptive immunity by leveraging the unique biology of iNKT cells, which can mediate direct cytotoxicity against tumor cells and orchestrate immune responses by targeting CD1d and other ligands in the tumor microenvironment. Pembrolizumab blocks the PD-1 receptor, releasing inhibitory signals on T-cells and promoting anti-tumor immunity. The combination is intended to enhance anti-tumor immune responses, particularly in tumors refractory to PD-1 blockade alone. The AGENT-797 + pembrolizumab combination has demonstrated early signals of clinical activity and a favorable safety profile in phase 1 clinical trials involving patients with advanced solid tumors, including those previously treated with other therapies[3][5]. AGENT-797 is manufactured as an off-the-shelf, HLA-unmatched therapy that does not require lymphodepletion[4].
