This is an experimental five-drug immunotherapeutic combination designed to enhance antitumor immunity through multiple synergistic mechanisms. It consists of a **CD40 agonist antibody** that activates dendritic cells by mimicking CD40 ligand binding to promote antigen presentation and T-cell priming; an **anti-CTLA4 antibody** that blocks cytotoxic T-lymphocyte-associated protein 4 to relieve T-cell inhibition and boost effector responses; a **STING agonist** that stimulates the stimulator of interferon genes pathway to induce type I interferons and innate immune activation; **resiquimod**, a small-molecule Toll-like receptor 7/8 agonist that drives proinflammatory cytokine production and myeloid cell maturation; and **interleukin-12 (IL-12)**, a cytokine that promotes Th1 differentiation, NK and CD8+ T-cell cytotoxicity, and IFN-γ secretion. The combination aims to remodel the immunosuppressive tumor microenvironment, as demonstrated in preclinical models where it elicited robust tumor regression.[1]
