**AGS-1C4D4 + gemcitabine** is a combination therapy for advanced/metastatic pancreatic cancer. AGS-1C4D4 is a fully human IgG1 monoclonal antibody that targets the prostate stem cell antigen (PSCA), a cell surface protein overexpressed in several cancers including pancreatic cancer. By binding to PSCA, AGS-1C4D4 induces complement-dependent cytotoxicity (CDC) and antibody-dependent cellular cytotoxicity (ADCC) in PSCA-expressing tumor cells. Gemcitabine is a nucleoside analog antineoplastic agent that inhibits DNA synthesis and induces apoptosis in dividing cells. This combination was evaluated in a phase II clinical trial, where the addition of AGS-1C4D4 to gemcitabine improved six-month survival and median overall survival versus gemcitabine alone in metastatic pancreatic adenocarcinoma patients[1][3][4][6].
