AGX148 + PH-762 is an investigational combination cell therapy under phase 1 clinical development for advanced solid tumors and melanoma. AGX148 refers to autologous “double positive” (CD39+ CD103+) CD8 tumor-infiltrating lymphocytes (TIL) isolated and expanded from solid tumors. PH-762 is a self-delivering small interfering RNA (siRNA) that silences PD-1 (programmed cell death protein 1) expression, designed to reduce T-cell exhaustion and enhance antitumor cytotoxicity. In this combination, AGX148 TILs are treated ex vivo with PH-762 prior to adoptive transfer in patients. Preclinical and early clinical data show that PH-762 treatment increases the tumor killing activity of AGX148 TILs approximately twofold, by inhibiting the PD-1 checkpoint, leading to greater T-cell-mediated tumor regression. The product is being developed by AgonOx and Phio Pharmaceuticals, and trials are run in collaboration with Providence Cancer Institute[1][2][3][4][5][6][7].
